Kidney disease, bone fractures, and more linked to PPIs

Research on the prolonged use of Proton Pump Inhibitors, like Dexilant, was recently published by the scholarly website JAMA showing a significant increased in the risk of developing chronic kidney disease (CKD):

In a prospective community-based cohort of more than 10 000 adults, we found that baseline use of PPIs was independently associated with a 20% to 50% higher risk of incident CKD.

In contrast to the recent evidence supporting the connection between heartburn drugs and kidney disease, PPIs have long been suspected of another severe side effect. Bone fractures have been reported in many claims against this class medications. In early 2010 the FDA released a notice indicating the requirement of PPI manufacturers to carry an additional warning notice regarding the risk of bone fracture. This notice was then updated to indicate that no such warning was required as the risk only applied to those using the medication for a prolonged period of time.

The side effects of Actos included congestive heart failure and bladder cancer. As a result, Takeda was forced to handle up to 10,000 bladder cancer lawsuits in the U.S. from those who were prescribed Actos.

Takeda eventually opted to settle the claims noting the following on their website:

Deerfield, Ill., U.S.,April 28, and Osaka, Japan, April 29, 2015 – Takeda Pharmaceutical Company Limited and its wholly-owned subsidiary, Takeda Pharmaceuticals U.S.A., Inc., today announced that they have reached an agreement expected to resolve the vast majority of ACTOS (pioglitazone HCl) product liability lawsuits pending against Takeda in the U.S. Takeda will take a $2.7 billion charge against earnings in the fourth quarter of fiscal year 2014 to cover the settlement and the costs associated with defending remaining cases and for other related litigation.

Dexilant, is yet another drug produced by Takeda, that is now linked dangerous side effects that were unknown when the drug was released to consumers.
